Client Overview:
Our client, a rapidly expanding biotechnology organization within a major U.S. biotech corridor, is seeking to advance their in vitro ADME and DMPK capabilities through automation and high-throughput innovation. They are currently looking to hire a Principal Scientist (High-Throughput ADME) to join their team. This organization is focused on modernizing ADME (Absorption, Distribution, Metabolism, and Excretion) platforms to accelerate small molecule drug discovery. By integrating high-throughput ADME screening, LC-MS/MS analytics, and automated laboratory systems, the company is transforming how ADME data informs medicinal chemistry and drug metabolism strategies.
Salary/Hourly Rate:
$75/hr - $90/hr
Position Overview:
The Principal Scientist (High-Throughput ADME) will lead the design and execution of advanced in vitro ADME assays to support drug metabolism and pharmacokinetics (DMPK) initiatives. The Principal Scientist (High-Throughput ADME) will play a critical role in scaling ADME screening platforms, delivering high-quality ADME data, and optimizing drug metabolism workflows to accelerate lead optimization. The Principal Scientist (High-Throughput ADME) position requires deep expertise in ADME assay development, LC-MS/MS analytics, and automated high-throughput screening technologies. The Principal Scientist (High-Throughput ADME) will collaborate closely with Medicinal Chemistry teams to ensure ADME and DMPK data drive strategic decision-making across the drug discovery pipeline. As a senior scientific leader, the Principal Scientist (High-Throughput ADME) will help modernize ADME infrastructure, enhance pharmacokinetics analysis, and strengthen in vitro ADME capabilities across multiple therapeutic programs.
Responsibilities of the Principal Scientist (High-Throughput ADME):
- High-Throughput ADME Assay Development & Automation:
- Design, develop, optimize, and validate high-throughput in vitro ADME assays, including solubility, MDCK permeability, plasma protein binding, microsomal stability, hepatocyte stability, and CYP inhibition/induction assays.
- Lead automation initiatives for ADME screening using liquid handling platforms such as Hamilton, Lynx, Bravo, and Formulatrix systems.
- Implement scalable workflows to enhance ADME throughput, DMPK data accuracy, and reproducibility across programs.
- Continuously refine ADME assay validation standards to support regulatory and scientific rigor.
- Drug Metabolism & LC-MS/MS Analysis:
- Develop and execute LC-MS/MS and acoustic mass spectrometry methods for in vitro ADME and drug metabolism studies.
- Oversee sample preparation, mass spectrometry method development, troubleshooting, and high-throughput ADME analysis.
- Analyze complex ADME and pharmacokinetics datasets, calculating intrinsic clearance, permeability coefficients, metabolic stability, and CYP inhibition parameters.
- Interpret ADME and DMPK data to guide medicinal chemistry optimization strategies.
- Present ADME findings and drug metabolism insights to cross-functional stakeholders.
- Quality, Compliance & Data Integrity:
- Maintain comprehensive documentation for ADME assays, DMPK workflows, SOPs, and electronic laboratory notebooks.
- Ensure all ADME and drug metabolism activities meet internal quality standards and laboratory safety regulations.
- Support data integrity for high-throughput ADME and pharmacokinetics reporting.
- Extensive hands-on experience with LC-MS/MS or acoustic mass spectrometry in high-throughput ADME environments.
- Proven track record in developing and executing automated in vitro ADME assays.
- Strong expertise in drug metabolism pathways, pharmacokinetics modeling, and DMPK strategy.
- Experience using ADME prediction software, pharmacokinetics tools, and data visualization platforms.
- Demonstrated experience scaling ADME screening operations in a biotech or pharmaceutical setting.
- Advanced laboratory automation expertise supporting ADME and DMPK workflows.
- Programming experience (Python, R, or similar) for ADME data analysis and pharmacokinetics modeling.
- Data visualization experience (Spotfire, Python, R) for ADME and drug metabolism reporting.
- Experience handling clinical samples in ADME or DMPK studies.
- Background working in or managing CRO relationships supporting ADME programs.
- Proficiency with LIMS, ELN, and electronic data capture systems for ADME and pharmacokinetics data management.
- Ph.D. in Pharmaceutical Sciences, Pharmacology, Analytical Chemistry, or a related field with 2+ years of industry experience in ADME and DMPK, OR
- Master's degree with 5+ years of relevant ADME, drug metabolism, or pharmacokinetics experience.
