The successful candidate is expected to have extensive experience and expertise in structure elucidation of metabolites, drug metabolism, bioactivation including covalent protein binding studies, enzymology and translation capabilities to resolve ADMET issues in support of drug discovery. The individual is also expected to have practical experiences and expertise in the conduct of radiolabeled in vitro and in vivo metabolism studies, first-in-human cold metabolism and human mass balance studies to support drug development. Extensive experience in the conduct of mechanistic drug metabolism experiments is also required to resolve clinical PK and regulatory issues.Responsibilities include:
The individual will be responsible for conducting metabolite profiling and identification of assigned projects using data-dependent LC-UV/MS or LC-RAD/MS. Upon review of the data, the individual will be expected to identify issues, propose potential solutions and conduct mechanistic studies to solve compound-related biotransformation and disposition issues. The successful applicant is expected to develop and implement innovative LC-MS/MS based automated metabolite profiling and identification methodologies, as well as possess experience in software-driven data-mining of metabolites, metabolite scale-up and isolation for NMR based structural elucidation. In addition, hands-on troubleshooting of LC-RAD/MS instruments, review/translating drug metabolism data, supervision of electronic archiving of RAD and MS data and hands-on lab experiments in support of Janssen discovery and development projects is required. The successful candidate is expected to provide scientific leadership and contribute to staff development in a team of biotransformation scientists. The ability to collaborate in a multidisciplinary research environment to address and design out ADMET/PK issues is also required as well as representation of the department on project teams involving the presentation of data at internal company meetings as well as external conferences.

Demonstrated skills and proficiency in all aspects of using nominal and high-resolution LC-MS/MS for automated metabolite profiling and identification of small organic drug molecules is required
Strong computer skills and experience with software driven metabolite profiling is required.
Detailed structure elucidation by NMR is preferred.
A strong background in interfacing separation techniques (2D, on-line SPE, and micro/nano LC) of drugs and its metabolites with mass spectrometry is preferred.
In-depth understanding of drug metabolism, enzymology, bioactivation, ADMET, and translating ADMET data to resolve DMPK issues is required.
Experience in metabolism of peptide and oligonucleotide drugs, and knowledgeable in protein and oligonucleotide chemistries is preferred.
Ability to independently diagnose and solve complex technical and scientific problems is required.
Self-motivated with excellent communication and interpersonal skills is required.
Able to effectively interface with others in a matrix team environment is required.
